*IMPORTANT!!!* Don't forget to make Kiddush on Yom Kippur

Making Kiddush on Yom Kippur is a מחלוקת between

ר' עקיבא איגר 

who holds it is only a מצוה דרבנן unless it falls
on a Shabbos and the 

חתם סופר 

(הגהות O.Ch.271) who 
holds it is a מצוה דאורייתא to make Kiddush every 
Yom Kippur. (even weekdays)

If it is a מצוה דאורייתא, one must have כוונה  in the 
שמונה עשרהi on Yom Kippur night when saying
ברוך אתה ה' – מקדש ישראל ויום הכפורים
to be מקיים  (Yotzei)  מצות קידוש.
שבת יו"כ everybody agrees that it is
a מצוה דאורייתא and one needs to have כוונה to
be יוצא
As far as I know, ספרי הלכה don't mention the מצוות קידוש ביו",כ
It is unfortunate how many people are unaware of this מצוה and
are not יוצא קידוש on Yom Kippur.

If one is unaware of this Halacha that מקדש ישראל ויום הכפורים
is the קידוש of Yom Kippur then he is a מתעסק (when saying it) 

Mitzvos דרבנן one is יוצא even without כוונה  However,
if one is מתעסק he is not יוצא even מצוות דרבנן.
Therefore one must at least know that the Bracha of מקדש ישראל
is the קידוש ליום כיפורים
According to the חתם סופר, or on שבת יו"כ  knowing is not enough
you must also have כוונה  to be יוצא קידוש when saying the Bracha
of מקדש ישראל.
